Application
Monitor transcriptional activity of progesterone receptor signaling pathway in mammalian cells, screen for activators or inhibitors of progesterone signaling pathway. The fluorescent reporter enables convenient readout using flow cytometry, fluorescence microscopy, etc. The progesterone response element (PRE) reporter lentivirus is constructed with tandem repeats of the consensus progesterone receptor DNA-binding element which is transcriptionally responsive to progesterone signaling pathway activity in mammalian cells. The physiological effects of progesterone are primarily mediated through progesterone receptor (PR) activation followed by receptor binding to a specific PRE, resulting in the transcription of target genes. Compared to PR-A, isoform PR-B is a transcriptional activator. The PR-B activation is stimulated by activation of MAPK-ERK pathway in the presence of its ligand progesterone.
