Application
Monitor PPAR-alpha activity, test for inhibitors of PPAR-alpha-mediated transcriptional activity. The PPARα reporter construct was optimized to allow for preferential activation by PPARα and RXRα heterodimers within peroxisome proliferator receptor elements (PPRE) in the genes regulated by PPARα. Tandem repeats of PPRE element on the Acox1 promoter of PPARα target gene can read out its transcriptional activity better than PPARγ in mammalian cells. PPARα is expressed highly in the liver and functions to control lipid metabolism, especially fatty acid oxidation.
