Monitor transcriptional activity of GATA3 in mammalian cells, screen for activators or inhibitors of GATA3 transcriptional activity. The fluorescent reporter enables convenient readout using flow cytometry, fluorescence microscopy, etc. The GATA3 Reporter Lentivirus is constructed with tandem repeats of the consensus GATA3 DNA-binding elements which is transcriptionally responsive to the transcription factor GATA3. GATA3 plays an essential role in the transcriptional control of Th2 cell differentiation and T lymphoid development and immune regulation. GATA3 is also expressed in multipotent hematopoietic stem cells and regulates the balance between self-renewal and differentiation. GATA proteins GATA1,2,3 have overlapping but distinct sequence preferences for DNA binding affinity. For example, GATA2 has been shown to bind the GATA3 binding motif which can only be poorly bound by GATA1. GATA1 and GATA2 play essential roles in the development and maintenance of the erythroid and megakaryocytic lineage, and the proliferation and survival of early hematopoietic cells, respectively.
